At the request of Monsigneur André Mulch, Archbishop of Rouen, Pope Paul VI decided on 6 July 1974 through the papal bull Quae Sacrosanctum on the creation of the diocese of Le Havre (Portus Gratiae in Latin meaning "Port of Grace"). The diocese was created from part of the parishes of the Archdiocese of Rouen to the west of a line joining Norville to Sassetot-le-Mauconduit. Monseigneur Michel Saudreau, its first bishop, was ordained on 22 September 1974. The church of Notre Dame was promoted to Cathedral Notre Dame du Havre. Today, the commune of Le Havre is divided into eight parishes and 24 places of worship (churches and chapels). The oldest chapel is Saint-Michel d'Ingouville which dates back to the 11th century. The Church of Saint Joseph du Havre, built by Auguste Perret, dominates the city with its spire 107m high. There are several monastic establishments (Carmel of the Transfiguration, Franciscan Monastery, Little Sisters of the Poor, etc.). The Protestant Church of Le Havre was built in the city centre in 1862. Bombed in 1941, it lost its pediment, its bell tower, and roof. Rebuilt in 1953 by the architects Jacques Lamy and Gérard Dupasquier, who worked in the Auguste Perret office, is the only building in Le Havre uniting the original architecture of the 19th century with the architecture of the Perret school. == Blunt-end ligation == Blunt end ligation does not involve base-pairing of the protruding ends, so any blunt end may be ligated to another blunt end. Blunt ends may be generated by restriction enzymes such as SmaI and EcoRV. A major advantage of blunt-end cloning is that the desired insert does not require any restriction sites in its sequence as blunt-ends are usually generated in a PCR, and the PCR generated blunt-ended DNA fragment may then be ligated into a blunt-ended vector generated from restriction digest. Blunt-end ligation, however, is much less efficient than sticky end ligation, typically the reaction is 100X slower than sticky-end ligation. Since blunt-end does not have protruding ends, the ligation reaction depends on random collisions between the blunt-ends and is consequently much less efficient. To compensate for the lower efficiency, the concentration of ligase used is higher than sticky end ligation (10x or more). The concentration of DNA used in blunt-end ligation is also higher to increase the likelihood of collisions between ends, and longer incubation time may also be used for blunt-end ligations. If both ends needed to be ligated into a vector are blunt-ended, then the vector needs to be dephosphorylated to minimize self-ligation. This may be done using CIAP, but caution in its use is necessary as noted previously. Since the vector has been dephosphorylated, and ligation requires the presence of a 5'-phosphate, the insert must be phosphorylated.
=== Continuous-wave (CW) spectroscopy === In the first few decades of nuclear magnetic resonance, spectrometers used a technique known as continuous-wave (CW) spectroscopy, where the transverse spin magnetization generated by a weak oscillating magnetic field is recorded as a function of the oscillation frequency or static field strength B0. When the oscillation frequency matches the nuclear resonance frequency, the transverse magnetization is maximized and a peak is observed in the spectrum. Although NMR spectra could be, and have been, obtained using a fixed constant magnetic field and sweeping the frequency of the oscillating magnetic field, it was more convenient to use a fixed frequency source and vary the current (and hence magnetic field) in an electromagnet to observe the resonant absorption signals. This is the origin of the counterintuitive, but still common, "high field" and "low field" terminology for low frequency and high frequency regions, respectively, of the NMR spectrum. As of 1996, CW instruments were still used for routine work because the older instruments were cheaper to maintain and operate, often operating at 60 MHz with correspondingly weaker (non-superconducting) electromagnets cooled with water rather than liquid helium. One radio coil operated continuously, sweeping through a range of frequencies, while another orthogonal coil, designed not to receive radiation from the transmitter, received signals from nuclei that reoriented in solution.
